A friend helped me set up my computer running XP pro. The harddrive is partitioned into an C: and D: configuration. However, when I try to access the D: drive, a message tells me the drive is not formatted. IT then tells me formatting will erase all data on the drive.

My question is this; if I format D:, will I lose the Data already on the C: portion of the drive? I am trying to figure out if I need to backup everything on my C: drive before I format.

No you would not lose any data on C if your formatted D, If you system files are on C then windows would not let you format C anyway, It's a very good ideal to do back ups anyway ,as i have learnt at great cost.
