Microphone Problems

Hello! I'm having problems with my microphone. It's a fully working mic, I even tested it on my other computer and it works but whenever i try to skype with friends on this computer, It will not let me speak. I've made sure the audio settings are correct and the microphone is connected. Is there a way to fix this?

Laptop or desktop?
How is the mic plugged in, USB or 3.5mm jack?
If its 3.5mm jack do you have headphones that you could plug into the 3.5mm jack for headphones to see if they work?

I can still hear from the headphones and its a turtle beach so it connects both usb and 3.5mm jack
